2015 a AtualInteração do Sistema Renina-Angiotensina com marcadores de inflamação e fibrose na progressão da Doença Renal Crônica
A DRC é considerada um dos mais importantes problemas de saúde pública mundial da atualidade. Apesar de a população pediátrica representar menor número de pacientes com DRC em relação à população adulta, a abordagem da DRC neste grupo se torna um desafio, por apresentar, além daquelas complicações comuns aos adultos, características únicas decorrentes das manifestações da doença em seres em crescimento e em desenvolvimento. O Sistema Renina Angiotensin (SRA) está intimamente envolvido nos mecanismos de progressão da lesão renal. Vários estudos clínicos mostram que a inibição da formação (IECA) e/ou da ligação da Ang II a seu receptor AT1 (ARA) pode retardar a progressão da fibrose renal e reduzir a mortalidade e o risco de complicações cardiovasculares relacionados à DRC. Nesta proposta, objetivamos investigar o papel do SRA na progressão da DRC e avaliar sua interação com moléculas imuno-inflamatórias e fibrogênicas. Serão também investigados como os componentes do SRA e as moléculas imuno-inflamatórias e fibrogênicas se associam às alterações clínicas, metabólicas, cardiovasculares e psiquiátricas das crianças e adolescentes com DRC, no intuito de identificar biomarcadores para diagnóstico e prognóstico bem como novos alvos terapêuticos. Esta proposta está claramente alinhada com os objetivos específicos do edital CNPq/MS/SCTIE/DECIT Nº 36/2014, sobretudo: ?Desenvolver pesquisas sobre os mecanismos básicos de doenças renais? e ?Investigar novos procedimentos diagnósticos e terapêuticos para doenças renais?. O projeto inclui pacientes pediátricos com DRC em diferentes estágios e de diferentes etiologias (abordagem clínica), além de modelos experimentais de doença renal (abordagem experimental). Os pacientes serão submetidos à avaliação clínica e coleta de amostras biológicas (sangue e urina) para mensurar componentes do SRA, moléculas imuno-inflamatórias, relacionadas à hemostasia, marcadores de metabolismo mineral ósseo e fibrogênicas, no intuito de identificar potenciais biomarcadores de diagnóstico, progressão da doença e resposta ao tratamento em pacientes com DRC. Os modelos experimentais de doenças renais (diferentes níveis de nefrectomia e nefropatia induzida pelo doxorrubicina) serão utilizados para investigar a fisiopatologia da lesão renal, os mediadores e vias metabólicas relacionados à piora da função renal e o papel da modulação do SRA sobre os processos de inflamação e fibrose renal. Além disso, esses modelos permitirão a investigação dos mecanismos de renoproteção de moléculas agonistas do receptor Mas e dos fármacos bloqueadores da via clássica do SRA. Pretende-se com o desenvolvimento do projeto aumentar os conhecimentos sobre a fisiopatologia da DRC.

1994 a AtualEstudos sobre as ações renais da Angiotensina-(1-7)
Trata-se de projeto iniciado a partir de minha dissertação de mestrado que envolveu o estudo das ações renais de um metabólito do Sistema Renina Angiotensina, a Angiotensina-(1-7), utilizando como ferramenta seu antagonista seletivo, o composto A-779. A partir daí os estudos vem avançando através do entendimento do papel da Angiotensina-(1-7) no controle do equilíbrio hidroeletrolítico e da descoberta de seu possível receptor, o receptor Mas.
